Prussian Blue Method for Hemosiderin

Fixation:
10% Buffered Neutral Formalin (#15740).
Sections:
Paraffin @ 5 microns
Staining Procedures:
- Deparaffinize and hydrate to distilled water.
- Place sections in working staining solution* for 30 minutes @ 60°C or for 1 hour at room temperature. Rinse in distilled water.
- Prepare working stain by mixing equal volumes of Potassium Ferrocyanide, 2% Aqueous (#26613-01) and Hydrochoric Acid, 2% (#26613-02). Prepare fresh just before use.
- Counter-stain in Safranin O, 0.2% (#26613-03) for 2 minutes. Wash in Acetic Acid, 1% (#26613-04)
- Dehydrate in 95% Alcohol, absolute alcohol, and clear in xylene, two changes each.
- Mount with Permount (#17986-01) or any synthetic resin.
